Gale, Glen R. (Veterans Administration Hospital and Duke University Medical Center, Durham, N.C.). Effects of the antifungal peptide Ro 2-7758 on morphology of Mucor corymbifera. J. Bacteriol. 85:833-837. 1963.-Hyphae of Mucor corymbifera grown in the presence of the antifungal peptide, Ro 2-7758, were examined in an electron microscope. The predominant change, compared with control hyphae, was a marked thickening of the cell wall, with apparent maintenance of normal structure. Cell walls of control hyphae were from 0.03 to 0.11 mu in thickness; drug-treated walls varied from 0.22 to over 0.75 mu in thickness. The possible sites and mechanisms of action, as well as the significance of a thickened cell wall in antibiosis, are discussed.

Multicenter evaluation of MIC distributions for epidemiologic cutoff value definition to detect amphotericin B, posaconazole, and itraconazole resistance among the most clinically relevant species of Mucorales.

Clinical breakpoints (CBPs) have not been established for the Mucorales and any antifungal agent. In lieu of CBPs, epidemiologic cutoff values (ECVs) are proposed for amphotericin B, posaconazole, and itraconazole and four Mucorales species. Wild-type (WT) MIC distributions (organisms in a species-drug combination with no detectable acquired resistance mechanisms) were defined with available po...

Multi-function Plant Defensin, Antimicrobial and Heavy Metal Adsorbent Peptide

Background: Defensin peptide isolated from plants are often heterogeneous in length, sequence and structure, but they are mostly small, cationic and amphipathic. Plant defensins exhibit broad-spectrum antibacterial and antifungal activities against Gram-positive and Gram-negative bacteria, fungi and etc. Plant defensins also play an important role in innate immunity, such as he...

In vitro interaction of posaconazole with calcineurin inhibitors and sirolimus against zygomycetes.

OBJECTIVES Zygomycosis is an uncommon but devastating disease with few therapeutic options. Calcineurin inhibitors and sirolimus (mTOR inhibitor), commonly used in transplant patients as immunosuppressives, have antifungal activity. They are known to demonstrate synergy with triazoles against certain fungi, though limited data exist about their activity against zygomycetes. Our aim was to study...

Evaluation of the post-antifungal effect (PAFE) of amphotericin B and nystatin against 30 zygomycetes using two different media.

The post-antifungal effect (PAFE) of amphotericin B and nystatin against 30 clinical zygomycetes was evaluated using two different media. PAFE is a suppression of fungal growth after limited drug exposure. The MICs of both drugs were determined using NCCLS M38-P guidelines. A spectrophotometric method was used to determine PAFE in vitro. Spores were exposed to amphotericin B and nystatin in RPM...

In Vitro Activity of Isavuconazole and Comparators against Clinical Isolates of the Mucorales Order

The in vitro activity of isavuconazole against Mucorales isolates measured by EUCAST E.Def 9.2 and CLSI M38-A2 methodologies was investigated in comparison with those of amphotericin B, posaconazole, and voriconazole. Seventy-two isolates were included: 12 of Lichtheimia corymbifera, 5 of Lichtheimia ramosa, 5 of group I and 9 of group II of Mucor circinelloides, 9 of Rhizomucor pusillus, 26 of...
